目的 了解托吡酯对兴奋性谷氨酸和癫发作引起神经元损伤的保护作用。方法 将不同浓度的托吡酯和谷氨酸作用于神经元后 ,观察神经元存活率的变化 ;测定服用托吡酯的病人在用药前后血中神经元烯醇化酶含量。结果 在 50 μmol/L、10 0μmol/L谷氨酸条件下 ,50~10 0 0 μmol/L托吡酯使神经元存活率分别由 30 9%、2 2 2 %增加到 4 6 9%~ 71 9%、4 0 4 %~ 6 8 1% (P <0 0 5)。先予托吡酯后加谷氨酸钠对神经元的保护作用优于先予谷氨酸钠后加托吡酯。服用托吡酯的病人在癫发作后血中神经元烯醇化酶量较服药前降低。结论 托吡酯对谷氨酸钠和癫发作引起的神经元死亡有保护作用
Objective To understand the protective effects of topiramate on neuronal damage caused by excitatory glutamate and epileptic seizures. Methods After different concentrations of topiramate and glutamic acid were applied to neurons, the change of neuronal survival rate was observed. The levels of neuron enolase in blood were measured before and after taking topiramate. Results Under 50 μmol / L and 100 μmol / L glutamic acid, the viability of neurons in 50 ~ 100 μmol / L topiramate increased from 30 9% and 22 2% to 46 9% ~ 71 9, respectively %, 4 0 4% ~ 6 8 1% (P <0 05). Topiramate plus monosodium glutamate after the protective effect of neurons is better than the first sodium glutamate after topiramate. Patients taking topiramate in the epileptic seizures after blood neuron enolase decreased compared with before taking. Conclusion Topiramate has a protective effect on the death of neurons caused by sodium glutamate and epileptic seizures
