目的分析引起吉林省2009年手足口病(Hand Food and Mouth Disease,HFMD)流行的肠道病毒71型(Enterovirus Type 71,EV71)分离株的基因特征。方法选择15株2009年分离于吉林省六个设区的市(州)的HFMD病例的EV71分离株,进行VP1编码区基因全长逆转录-聚合酶链反应扩增、序列测定,使用Bioedit软件和MEGA4.0软件进行同源性、基因亲缘关系分析。结果吉林省2009年的15株EV71分离株均属于C4基因亚型,而且分属于四个相对独立的传播链,其中11株病毒(分离于六个市)属于一个较大的传播链,其余4株形成三个小的传播链。29.5%的吉林省分离株间比对同源性高达99.1%~99.8%,吉林省2009年分离株与安徽省阜阳市2008年以及北京市2008年的流行株间,核苷酸最高同源性分别达到了99.8%和99.7%。结论吉林省2009年至少有四个C4基因型的EV71传播链引起了HFMD的流行,并且有一个EV71优势传播链在几个省以及省内六个市持续传播,引起HFMD的流行;三个小的传播链可能为输入病毒所致。尚未发现在引起HFMD死亡、重症以及轻症的EV71分离株VP1编码区的氨基酸序列呈现规律性的区别。
Objective To analyze the genetic characteristics of Enterovirus Type 71 (EV71) isolates that caused the 2009 Hand Food and Mouth Disease (HFMD) epidemic in Jilin Province. Methods Fifteen EV71 isolates from HFMD cases in the six prefectures of Jilin Province in 2009 were selected and amplified by full length RT - PCR and sequenced using Bioedit software And MEGA4.0 software homology, genetic relationship analysis. Results In 2009, all the 15 EV71 isolates belonged to the C4 gene subtype and belonged to four relatively independent transmission chains, of which 11 strains (separated from the six cities) belonged to a larger transmission chain and the remaining 4 Strains form three small transmission chains. The homology of 29.5% isolates in Jilin Province was as high as 99.1% ~ 99.8% among the isolates in Jilin Province in 2009, Fuyang City in Anhui Province in 2008 and the epidemic strains in Beijing in 2008. Conclusions In 2009, at least four EV71 transmission lines of C4 genotype caused the epidemic of HFMD in Jilin Province and one EV71 dominant transmission line continued to spread in several provinces and six provinces in the province, causing the epidemic of HFMD.
