目的:探讨藏药湿生扁蕾对人结肠癌细胞SW480增殖调控因子CyclinD1、CDK4、PCNA和p21kip1·mRNA表达的影响。方法:将SW480细胞接种于6孔培养板,24h后分别用不同浓度(0.25、0.50、1.00 mg/ml)的湿生扁蕾干预,继续培养24h,提取每组总RNA,以GAPDH基因为内参,采用RT-PCR法检测CyclinD1、CDK4、PCNA及p21kip1的mRNA表达水平。结果:PCNA、CyclinD1及CDK4的mRNA表达随湿生扁蕾浓度增加而减少,而p21kip1mRNA的表达随湿生扁蕾浓度增加而增加,并呈现明显的剂量依赖关系。结论:藏药湿生扁蕾干预SW480细胞的增殖与其抑制CyclinD1、CDK4、PCNA的表达和促进p21kip1的表达作用有关。
OBJECTIVE: To investigate the effect of Tibetan hyoid flower bud on the expression of CyclinD1, CDK4, PCNA and p21kip1 · mRNA in human colon cancer cell line SW480. Methods: SW480 cells were seeded on a 6-well plate. After 24 hours, the cells were cultured for 24 hours with different concentrations (0.25, 0.50 and 1.00 mg / ml) respectively. Total RNA was extracted from each group. GAPDH gene was used as an internal control The mRNA expression of CyclinD1, CDK4, PCNA and p21kip1 were detected by RT-PCR. Results: The mRNA expressions of PCNA, CyclinD1 and CDK4 decreased with the increase of Hygrophylla spp., While the expression of p21kip1 mRNA increased with the increase of hygrophytiaphyla spp., And showed a significant dose - dependent relationship. Conclusion: The proliferation of SW480 cells induced by Tibetan hypospaditis is related to the inhibition of the expression of CyclinD1, CDK4 and PCNA and the expression of p21kip1.
