目的探讨NUCB2/Nesfatin-1在饮食诱导肥胖(DIO)大鼠下丘脑中的表达水平,为糖脂代谢神经调节机制的深入研究提供实验依据。方法通过高能饲料喂养建立高血糖高血脂大鼠模型,采用免疫组织化学方法检测下丘脑NUCB2/Nesfatin-1表达水平。结果 1NUCB2/Nesfatin-1在实验组大鼠下丘脑表达的阳性率为43.33%,明显低于对照组的80.00%(P<0.05)。2 NUCB2/Nesfatin-1在实验组大鼠下丘脑的表达强度积分为2.159±0.918,对照组表达积分为3.683±1.250(P<0.01)。结论 NUCB2/Nesfatin-1在饮食诱导肥胖(DIO)大鼠下丘脑的表达水平明显降低,提示NUCB2/Nesfatin-1可能在糖脂代谢神经调节中具有重要作用。
Objective To investigate the expression of NUCB2 / Nesfatin-1 in the hypothalamus of diet-induced obesity (DIO) rats and provide experimental evidence for the further study on the neuromodulation mechanism of glycolipid metabolism. Methods Hyperglycemia and hyperlipidemia rats were established by feeding with high energy diet. The expression of NUCB2 / Nesfatin-1 in the hypothalamus was detected by immunohistochemistry. Results The positive rate of 1NUCB2 / Nesfatin-1 expression in the hypothalamus of the experimental group was 43.33%, which was significantly lower than that of the control group (80.00%, P <0.05). The expression of NUCB2 / Nesfatin-1 in the hypothalamus of experimental group was 2.159 ± 0.918, and that in the control group was 3.683 ± 1.250 (P <0.01). Conclusions The expression level of NUCB2 / Nesfatin-1 in the hypothalamus of diet-induced obesity (DIO) rats is obviously decreased, suggesting that NUCB2 / Nesfatin-1 may play an important role in the neurotransmission of glucose and lipid metabolism.
