目的 使用脂质体介导的端粒酶反义寡核苷酸封闭端粒酶基因在视网膜色素上皮细胞的表达从而抑制视网膜色素上皮细胞的增殖,为增殖性玻璃体视网膜病变的治疗探索一条基因治疗途径。方法 使用端粒重复序列扩增法(Telomeric repeat amplification protocol, TRAP)定性、定量检测浓度为Oumol/L、2umol/L、4umol/L、6umol/L、8umol/L脂质体介导的端粒酶反义寡核苷酸及浓度为4umol/L的正义寡核苷酸在视网膜色素上皮细胞端粒酶活性表达。结果 TRAP结果显示随着端粒酶反义寡核苷酸浓度的增高,视网膜色素上皮细胞端粒酶活性逐渐降低,8umol/L几乎没有表达,正义寡核昔酸很少抑制视网膜色素上皮细胞端粒酶活性。结论 脂质体介导的瑞粒酶反义寡核苷酸能抑制视网膜色素上皮细胞端粒酶的活性从而抑制视网膜色素上皮细胞的增殖。
OBJECTIVE: To investigate the gene therapy of proliferative vitreoretinopathy by using liposome-mediated telomerase antisense oligonucleotide to block the expression of telomerase gene in retinal pigment epithelial cells and thereby inhibit the proliferation of retinal pigment epithelial cells way. Methods Telomere repeat amplification protocol (TRAP) was used to characterize the telomeres mediated by liposomes at concentrations of 0, 2, 4, 6, 8 μmol / L Enzyme antisense oligonucleotides and sense oligosaccharides with a concentration of 4 umol / L were expressed in retinal pigment epithelial cells with telomerase activity. Results The results of TRAP showed that the telomerase activity of retinal pigment epithelial cells decreased gradually with the increase of telomerase antisense oligonucleotide concentration and almost no expression was found at 8umol / L, and the sense oligodeoxynucleotides seldom inhibited retinal pigment epithelium Granzyme activity. Conclusion Liposome-mediated resveratrol antisense oligonucleotide can inhibit the activity of telomerase in retinal pigment epithelial cells and inhibit the proliferation of retinal pigment epithelial cells.
