目的:探讨中药复方柴郁地仙方治疗围绝经期抑郁症(PDD)的作用机制。方法:48只3~4月龄雌性SD大鼠随机分为假手术组、模型组、西药组及柴郁地仙方高、中、低剂量组,每组8只。假手术组不予去势,其余组行卵巢去势术后孤养并予以28 d慢性不可预见性应激建立PDD模型,造模同时各组分别予柴郁地仙方、盐酸氟西汀和戊酸雌二醇混悬液及0.9%Na Cl溶液连续灌胃28 d。观察应激过程中大鼠行为学及体质量变化;采用酶联免疫吸附测定法(ELISA)检测血清中促肾上腺皮质激素释放激素(CRH)、促肾上腺皮质激素(ACTH)、皮质酮(CORT)含量。结果:模型组大鼠体质量增长缓慢,旷场水平得分及糖水偏好率相对于假手术组均明显降低(P<0.01);血清中CRH、ACTH、CORT含量较假手术组明显升高(P<0.01)。与模型组比较,柴郁地仙方高、中、低剂量组大鼠旷场水平得分、糖水偏好率及体质量均明显升高(P<0.01),血清中的CRH、ACTH、CORT含量明显降低(P<0.05,P<0.01)。结论:柴郁地仙方可能通过下丘脑-垂体-肾上腺轴的调节作用而起到治疗围绝经期抑郁症的作用。
Objective: To investigate the mechanism of traditional Chinese medicine compound Chai Yu Di Xian Fang in the treatment of perimenopausal depression (PDD). Methods: Forty-eight female SD rats of 3 to 4 months old were randomly divided into sham-operation group, model group, western medicine group and Chaiyudi Xian Fang high, medium and low dose group, 8 rats in each group. The sham-operation group was not castrate, the other group ovariectomized and isolated and 28d chronic unpredictable stress to establish PDD model, while the model was given to each group of Chai Yu Di Xian Fang, fluoxetine hydrochloride and Estradiol valerate suspension and 0.9% NaCl solution for 28 days. The changes of behavior and body weight of rats during the stress were observed. Serum corticotropin-releasing hormone (CRH), adrenocorticotropic hormone (ACTH) and corticosterone (CORT) were detected by enzyme linked immunosorbent assay (ELISA) content. Results: The body weight of rats in the model group increased slowly, the scores of open field and the preference of sugar were significantly lower than those in the sham operation group (P <0.01), while the contents of CRH, ACTH and CORT in the model group were significantly higher than those in the sham operation group <0.01). Compared with the model group, the scores of open field, preference rate of carbohydrate and body weight of rats in high, medium and low doses of Chaiyu were significantly higher (P <0.01), and the contents of CRH, ACTH and CORT in serum were significantly higher Decreased (P <0.05, P <0.01). Conclusion: Chai Yu Di Xian Fang may play a role in the treatment of perimenopausal depression by regulating the hypothalamic-pituitary-adrenal axis.
