目的探讨莪术油对体外培养的人乳腺癌MCF-7细胞增殖及诱导凋亡的作用。方法莪术油不同浓度(150、200、250、300和400mg·L-1)处理MCF-7细胞至对数生长期。MTT法筛选最佳药物处理浓度和时间的组合,计算抑制率。流式细胞技术检测MCF-7细胞的凋亡,并计算凋亡指数。Western blot印迹法及免疫组化法分别检测各处理组MCF-7细胞中Fas/FasL蛋白的表达。结果莪术油对MCF-7细胞增殖的抑制率随浓度的增加而增高(P<0.05),莪术油诱导MCF-7细胞凋亡率较对照组显著增加(P<0.01),且Fas蛋白表达增加(P<0.05),FasL蛋白表达减弱(P<0.05)。结论莪术油可抑制乳腺癌MCF-7细胞增殖,促进细胞凋亡,其机制可能与上调Fas蛋白、下调FasL蛋白表达有关。
Objective To investigate the effects of zedoary turmeric oil on the proliferation and apoptosis of human breast cancer cell line MCF-7 cultured in vitro. Methods Curcuma oil treated MCF-7 cells at various concentrations (150, 200, 250, 300 and 400 mg · L -1) to logarithmic growth phase. MTT method screening the best drug treatment concentration and time combination to calculate the inhibition rate. Flow cytometry was used to detect the apoptosis of MCF-7 cells and the apoptotic index was calculated. Western blotting and immunohistochemistry were used to detect the expression of Fas / FasL protein in MCF-7 cells. Results The inhibition rate of Curcuma oil on MCF-7 cells proliferation increased with the increase of concentration (P <0.05), and the apoptosis rate of MCF-7 cells increased significantly (P <0.01) and the expression of Fas protein increased (P <0.05), FasL protein expression decreased (P <0.05). Conclusion Curcuma oil can inhibit the proliferation and promote the apoptosis of MCF-7 breast cancer cells. The mechanism may be related to the up-regulation of Fas protein and down-regulation of FasL protein expression.
