目的探讨人白血病骨髓间充质干细胞(LMSC)对K562细胞生长曲线及细胞周期分布的影响。方法采用噻唑蓝(MTT)法,绘制与LMSC共培养前后K562细胞的生长曲线;流式细胞仪检测与LMSC共培养前后,K562细胞周期的变化。结果与LMSC共培养后,K562细胞生长受抑,G0~G1期细胞比例明显增高(47.3±1.3)%,S期细胞比例明显下降(39.1±0.6)%,二者与K562细胞单独悬浮培养组[G0~G1期:(39.6±1.2)%,S期:(48.0±1.2)%]相比,差异均有统计学意义(P<0.05);与LMSC共培养后,G2~M期细胞比例虽然也呈上升趋势,但差异无统计学意义(P>0.05)。结论①与LMSC共培养后K562细胞生长受抑制。②LMSC通过使K562细胞周期阻滞于G0~G1期发挥抑制K562细胞生长的作用。
Objective To investigate the effects of human leukemia bone marrow mesenchymal stem cells (LMSCs) on the growth curve and cell cycle distribution of K562 cells. Methods The growth curve of K562 cells before and after co-culture with LMSC was drawn by MTT method. The cell cycle of K562 cells was detected by flow cytometry before and after co-culture with LMSC. Results After co-cultured with LMSC, the growth of K562 cells was inhibited. The proportion of cells in G0-G1 phase was significantly increased (47.3 ± 1.3)%, and the percentage of cells in S phase was significantly decreased (39.1 ± 0.6)%. (P <0.05). Compared with LMSC group, the percentage of cells in G2 ~ M phase was significantly higher than that in control group [G0-G1 phase: (39.6 ± 1.2)%, S phase: (48.0 ± 1.2)%] Although it also showed an upward trend, the difference was not statistically significant (P> 0.05). Conclusion ① The growth of K562 cells was inhibited after co-cultured with LMSC. ② LMSC can inhibit the growth of K562 cells by blocking the cell cycle of K562 in G0 ~ G1 phase.
