目的评价滤纸片干血斑在HIV-1 BED-CEIA新发感染检测中洗脱液放置时间对结果判定的影响。方法使用2013年新发感染干血斑国际考核盘进行HIV-1 BED-CEIA检测,检验干血斑考核盘在BED-CEIA新发感染检测结果的稳定性与可重复性;洗脱液放置13 h、17 h、20 h、24 h、32 h后检测,进行随机区组设计方差分析。结果检测稳定性较好,6次检测质控品吸光度值(OD值)与标化吸光度(ODn值)的变异系数(CV)<15%;国际考核盘干血斑样8份,其不同放置时间5次检测判定为新发感染结果一致,即可重复性好;洗脱液放置13 h、17 h、20 h、24 h、32 h后检测,标化吸光度(ODn值)差异无统计学意义(χ2=0.705 7,P>0.05)。结论在HIV-1 BED-CEIA新发感染检测中,干血斑样本的检测结果具有较好的稳定性与可重复性,因此可以在今后检测工作中,应用干血斑样本进行HIV-1 BED-CEIA新发感染检测。
Objective To evaluate the impact of filter paper dried blood spots on the determination of results in the detection of HIV-1 BED-CEIA infection. Methods The BED-CEIA test of HIV-1 was performed using the 2013 international test of freshly infected dry blood spots to test the stability and repeatability of the test results of fresh blood spots in newly infected BED-CEIA. The eluent was placed 13 h, 17 h, 20 h, 24 h, 32 h after the randomized block design analysis of variance. Results The detection stability was good. The coefficient of variation (CV) <15% of absorbance value (OD value) and standard absorbance (ODn value) of 6 control samples were tested. The results of five times of tests showed that the new infection was consistent, and the reproducibility was good. After the elution was allowed to stand for 13 h, 17 h, 20 h, 24 h, 32 h, the normalized absorbance (ODn) showed no statistical difference Significance (χ2 = 0.705 7, P> 0.05). Conclusion In the detection of new HIV-1 BED-CEIA infection, the test results of dried blood spot samples have good stability and repeatability. Therefore, it is feasible to apply dried blood spot samples to HIV-1 BED -CEIA New infection detection.
