目的:建立妇乐胶囊中大黄素,大黄酚的含量测定方法。方法:采用高效液相色谱法,Shim-pack VP-ODS色谱柱(4.6 mm×250 mm,5μm),柱温30℃,流动相为甲醇-0.1%磷酸溶液(80:20),流速1.0 m L·min~(-1),检测波长254 nm,进样量10μL。结果:在上述色谱条件下,大黄素、大黄酚之间有良好的分离度,大黄素线性范围6.2784~125.5668 ng(r=0.9997),大黄酚线性范围10.76~215.28 ng(r=0.9999),精密度、重复性、稳定性的RSD均<2%,平均加样回收率分别为100.9%,99.6%,RSD分别为1.00%,0.71%。结论:所建方法操作简便、准确,重复性好,可作为妇乐胶囊的质量控制方法。
Objective: To establish the method of determination of emodin and chrysophanol in Fule capsule. Methods: High performance liquid chromatography (HPLC) was performed on a Shim-pack VP-ODS column (4.6 mm × 250 mm, 5 μm) with a column temperature of 30 ℃ and a mobile phase of methanol-0.1% phosphoric acid L · min ~ (-1), detection wavelength 254 nm, injection volume 10μL. Results: Under the above chromatographic conditions, there was good resolution between emodin and chrysophanol. The linear range of emodin was 6.2784 ~ 125.5668 ng (r = 0.9997). The linear range of chrysophanol was 10.76 ~ 215.28 ng (r = 0.9999) The RSDs of degree, repeatability and stability were all less than 2%. The average recoveries were 100.9% and 99.6% respectively. The RSDs were 1.00% and 0.71% respectively. Conclusion: The established method is simple, accurate and reproducible, and can be used as a quality control method for Fule capsule.
