目的运用多重PCR技术对食品中的致病菌检测,确认该方法在食源性微生物检测的适用性和有效性,为食品中致病菌的检测提供快速、准确、有效的方法与决策依据。方法在防城港市防城区和港口区采集具有代表性样品对7种等常见致病菌检验、分析和评估,同时与传统分离培养方法进行对照研究。结果 290份样品分别用2种方法进行检测,传统培养法检出致病菌82株,致病菌检出率为28.3%,其中PCR法检出138份,致病菌检出率为47.6%,两者检出率差异有统计学意义(P<0.05)。结论多重PCR法同样适合不同类型食品的检测,有较强的高效性,灵敏度高,操作简便,适合检测成组病原微生物,对食源性致病菌做出初步的判断,再采用传统的培养法有针对性地加以确认,极大减轻了工作量和提高了工作质量,对及时判定病原菌、有效控制病原菌传播、预防食物中毒的发生具有重要意义。
Objective To detect the pathogenic bacteria in food by multiplex PCR and to confirm the applicability and effectiveness of this method in the detection of foodborne microorganisms and to provide a fast, accurate and effective method and decision basis for the detection of pathogenic bacteria in food. Methods The representative samples were collected from Fangcheng District and the port area of Fangchenggang City to test, analyze and evaluate the seven common pathogenic bacteria. At the same time, they were compared with the traditional isolation and culture methods. Results 290 samples were detected by two methods, 82 strains of pathogens were detected by traditional culture method, the detection rate of pathogenic bacteria was 28.3%, of which 138 were detected by PCR method and the detection rate of pathogenic bacteria was 47.6% , The difference between the two detection rates was statistically significant (P <0.05). Conclusion Multiplex PCR method is also suitable for the detection of different types of food, with high efficiency, high sensitivity, simple operation, suitable for testing a group of pathogenic microorganisms, to make preliminary judgments on foodborne pathogens, and then using traditional culture Law to be targeted to confirm, greatly reducing the workload and improve the quality of work, timely identification of pathogens, effective control of the spread of pathogens, prevention of food poisoning is of great significance.
