目的:研究清心养神方药对应激大鼠心肌细胞肾上腺素β受体的干预效果。方法:采用放射配基结合法,分析实验心肌细胞肾上腺素β受体亲和力、密度和RT-PCR实验检测心肌细胞肾上腺素β受体mRNA,观察清心养神方药对束缚应激大鼠心肌细胞肾上腺素β受体变化的干预作用,并设心得安和朱砂安神丸组作为对照。结果:清心养神方药、心得安和朱砂安神丸均能增加心肌细胞肾上腺素β受体的亲和性,起到增敏现象;对应激引起的受体密度下降有明显抑制作用;对应激引起的β1-AR mRNA表达下调有抑制作用;可上调应激大鼠β2-AR mRNA表达。结论:清心养神方药具有类心得安样受体阻滞作用,对应激引起的心肌细胞肾上腺素β受体改变具有调节作用。
Objective: To study the effect of Qingxin Yangshen Prescription on the intervention of adrenaline β receptor in cardiomyocytes of stress rats. Methods: Radioimmunoassay method was used to analyze the adrenergic receptor β-adrenoceptor affinity, density and RT-PCR assay of cardiomyocyte adrenaline β receptor mRNA in experimental cardiomyocytes. The effect of qingxin recuperation prescription on myocardial cell adrenaline β receptor changes in the intervention, and set up peace of mind and cinnabar Anshen pill group as a control. Results: Qingxin recuperation prescription, propranolol and cinnabar and Anshen pills can increase the affinity of cardiomyocytes adrenaline β receptors, play a sensitization phenomenon; the stress caused by the receptor density decreased significantly inhibited; stress-induced β1-AR mRNA expression was inhibited; up-regulated β2-AR mRNA expression in rats. Conclusion: Qingxin Yangshen Recipe has therapies such as propranolol-like blockade and regulates cardiomyocyte adrenergic β-receptor changes induced by stress.
