目的:应用库尔特计数仪测定全氟丙烷人血白蛋白微球注射液中微球浓度和直径。方法:样品充分混匀后,移取20μl置于100 ml量瓶中,用0.9%氯化钠溶液定容,摇匀后立即测定。结果:样品稀释后微球浓度在2.94×10~4~29.45×10~4个/ml范围内线性关系良好(r=0.999 9),平均回收率为99.4%,RSD为1.95%,微球平均直径RSD为0.86%。结论:此方法简单、快速,结果准确可靠,可用于全氟丙烷人血白蛋白微球注射液中白蛋白微球的粒度测定。
Objective: To determine the concentration and diameter of microspheres in perfluoropropane human albumin microsphere injection by Coulter counter. Methods: After the sample was well mixed, remove 20μl into 100ml volumetric flask and dilute with 0.9% sodium chloride solution. Shake well and measure immediately. Results: The average concentration of microspheres in the range of 2.94 × 10 ~ 4 ~ 29.45 × 10 ~ 4 / ml was linear (r = 0.999 9), the average recovery was 99.4% and the RSD was 1.95% The diameter RSD is 0.86%. Conclusion: This method is simple, rapid, accurate and reliable. It can be used to determine the particle size of albumin microspheres in perfluoropropane human albumin microsphere injection.
