为了优化蜡梅叶片总黄酮含量的提取方法,本研究采用超声波提取,及以乙醇浓度、超声时间和超声温度为自变量,以总黄酮含量为响应值设计的响应面(RSM)优化方案。结果显示,以超声波及响应面法提取蜡梅叶片中总黄酮的最优提取方案为:乙醇浓度为85%,超声时间为34 min,超声温度为46℃,理论上预测的总黄酮含量为23.46 mg/g,测定的实际总黄酮含量为23.52±0.76 mg/g,模型的预测值与实际实验结果值相差很小,说明利用响应面法得到的提取参数比较可靠,可用于实际操作。另外,响应面法可以找出整个区域上因素的最佳组合和最优值,可信度较高。
In order to optimize the extraction method of total flavonoids content in Chimonanthus praecox, we optimized the response surface (RSM) using ultrasonic extraction and the concentration of ethanol, ultrasonic time and ultrasonic temperature as independent variables and the total flavonoid content as response value. The results showed that the optimal extraction conditions of total flavonoids from Chimonanthus praecox were: ethanol concentration 85%, sonication time 34 min, ultrasonic temperature 46 ℃, and the theoretically predicted total flavonoid content was 23.46 mg / g, the actual content of total flavonoids measured was 23.52 ± 0.76 mg / g. The predicted value of the model was not significantly different from the actual value of the experimental results, indicating that the extraction parameters obtained by the response surface method are more reliable and can be used for practical operation. In addition, the response surface method can find out the best combination and optimal value of the factors in the whole area, with higher credibility.
