赤芍承气汤对急性肝衰竭大鼠保护作用的实验研究

目的:探讨赤芍承气汤对内毒素脂多糖(LPS)致急性肝衰竭大鼠的保护作用。方法:以内毒素(LPS)加 D-氨基半乳糖(D-GalN)联合制备大鼠急性肝衰竭合并内毒素血症模型,以促肝细胞生成素颗粒剂(促肝康)和赤芍承气汤对模型动物进行治疗,观察血清 ALT、AST、TBIl 及肿瘤坏死因子-α(TNF-α)、内毒素(ET)的变化;常规 HE 染色,观察大鼠肝组织病理学改变;以免疫组化法检测 Fas、FasL 蛋白在大鼠肝组织中的表达情况,观察赤芍承气汤对肝细胞凋亡的作用。结果:LPS 加 D-GalN 可引起严重的急性肝坏死并有广泛的肝细胞凋亡。模型组 ALT、AST、TNF-α、ET 均显著升高,伴 Fas、FasL 蛋白在肝细胞中表达增强;中药组 ALT 及以上各指标的水平均较模型组明显降低(P<0.05),中药组大鼠肝坏死程度也较模型组明显减轻,肝细胞 Fas、FasL 阳性率,模型组为83%、92%,中药组为53%、50%,经统计学分析,差异有显著性(P<0.01)。促肝康可明显降低血清 ALT、AST,减轻肝组织病理损害。结论:赤芍承气汤能降低 ET、TNF-α的生成和释放,它和促肝康均具有良好的保护肝细胞、抗肝衰竭的作用;赤芍承气汤可减轻肝细胞 Fas/FasL 的表达,抵抗内毒素诱发的肝细胞凋亡。 Objective: To investigate the protective effect of Chikan Chengqi Decoction on rats with acute liver failure induced by endotoxin lipopolysaccharide (LPS). METHODS: Acute hepatic failure combined with endotoxemia model was induced by endotoxin (LPS) combined with D-galactosamine (D-GalN) to induce hepatocyte growth promoting granules (Zhugankang) and Akasaka. The model animals were treated with Tang, and the changes of serum ALT, AST, TBI1, tumor necrosis factor-α (TNF-α) and endotoxin (ET) were observed. After routine HE staining, the pathological changes of liver tissue were observed; The expression of Fas and FasL protein in rat liver tissue was detected by chemical method. The effect of Chiqi Chengqi Decoction on hepatocyte apoptosis was observed. Results: LPS plus D-GalN caused severe acute hepatic necrosis and extensive hepatocyte apoptosis. The levels of ALT, AST, TNF-α, and ET in the model group were significantly increased, and the expression of Fas and FasL protein was increased in hepatocytes. The levels of ALT and above indexes in the Chinese medicine group were significantly lower than those in the model group (P<0.05). The degree of hepatic necrosis in the rats was also significantly reduced compared with the model group. The positive rates of Fas and FasL in liver cells were 83% and 92% in the model group and 53% and 50% in the Chinese medicine group. Statistically significant differences were observed (P < 0.05). <0.01). Xugankang can significantly reduce serum ALT, AST and reduce liver pathological damage. Conclusion: Chiling Chengqi Decoction can reduce the production and release of ET and TNF-α. It and Hegankang have good protection of hepatocytes and anti-hepatic failure. Chiqi Chengqi Decoction can reduce hepatocyte Fas/FasL. The expression is resistant to endotoxin-induced hepatocyte apoptosis.
