目的探讨耐力训练对老龄大鼠铁代谢水平及铁调素(Hepcidin)mRNA表达的影响。方法选用SD雄性大鼠48只,随机分为安静对照组(NC组);低强度耐力组(LC组);中等强度耐力组(MC组);大强度耐力组(HC组),每组12只。5周耐力训练结束后,宰杀大鼠。根据试剂盒的方法测定老龄大鼠RBC、Hb、Hct、PI和铁蛋白含量并且通过逆转录聚合酶链式反应(RT-PCR)测定老龄大鼠肝组组织Hepcidin mRNA基因表达。结果老龄大鼠红细胞数含量随着运动强度的增加而出现下降的趋势;通过耐力训练后,血红蛋白含量在低强度、中等强度和大强度耐力训练低于安静对照组(P值分别为0.0082、0.0089、0.0053),红细胞比容在中等强度和大强度耐力训练时低于安静对照组(P值分别为0.0438、0.0413);在中等强度和大强度耐力训练时大鼠血清铁含量低于安静对照组(P值分别为0.0387、0.0401);铁蛋白含量在大强度耐力训练时低于安静对照组(P=0.0410)。肝组织Hepcidin mRNA表达量在低强度、中等强度和大强度耐力训练组高于安静对照组(P=0.0312、0.0327或P=0.0072)。结论耐力训练致使大鼠机体内铁代谢的缺乏和大鼠肝分泌的Hepcidin mRNA表达量出现上升的趋势。
Objective To investigate the effects of endurance training on iron metabolism and expression of Hepcidin mRNA in aged rats. Methods Forty-eight SD male rats were randomly divided into 4 groups: control group (NC group), low-intensity endurance group (LC group), moderate-intensity endurance group (MC group), high-intensity endurance group (HC group) only. After 5 weeks of endurance training, slaughtered rats. The contents of RBC, Hb, Hct, PI and ferritin in old rats were determined by kit method. Hepcidin mRNA expression in the liver of aged rats was determined by reverse transcription-polymerase chain reaction (RT-PCR). Results The content of erythrocytes in old rats tended to decline with the increase of exercise intensity. After endurance training, hemoglobin content was lower in low, moderate and high endurance training than in quiet control group (P = 0.0082, 0.0089 , 0.0053). The hematocrit was lower than that of the quiet control group (P = 0.0438, 0.0413, respectively) during moderate and intensive endurance training. Serum iron content was lower in the moderate and intensive endurance training rats than in the quiet control group (P = 0.0387, 0.0401, respectively). The content of ferritin was lower than that of the quiet control group during the intensive endurance training (P = 0.0410). Hepcidin mRNA expression in liver tissue was significantly higher in the low-intensity, moderate-intensity and high-intensity endurance training groups than in the quiet control group (P = 0.0312,0.0327 or P = 0.0072). Conclusions Endurance training results in the lack of iron metabolism and Hepcidin mRNA excretion in rat liver.
