目的 检查除草醚 (nitrofen)诱导的先天性膈疝大鼠动物模型气管及支气管是否异常。方法 大白鼠在妊娠 9.5d时经胃管注入 10 0mg的除草醚 ,妊娠 2 1d(预产期前 2 4h)行剖腹产 ,解剖胎鼠。其气管及支气管用Alcianblue alizarin染色 ,在外科显微镜下检查其脏器。其结果与 11只正常胎鼠对照。结果 正常对照胎鼠气管及支气管结构无异常 ,有先天性膈疝 (CDH)胎鼠气管环软骨数明显下降。 5 7只先天性膈疝胎鼠中 ,40只有不完全的气管软环 ,12只有程度不等的气管狭窄 ,并与异常的血管环相关。Nitrofen诱导而无先天性膈疝的胎鼠也有气管环数量减少 ,4只有轻微的气管狭窄。结论 Nitrofen诱导的大白鼠胎鼠除了有先天性膈疝合并肺发育不良 ,有时也会合并严重的气管支气管异常。提示在胚胎时其发病机理涉及到多种组织成分。致畸因子可能干涉了组织器官发育的调控基因。
Objective To investigate whether nitrofen - induced congenital diaphragmatic hernia in the trachea and bronchus of an animal model is abnormal. Methods Rats were injected with 100 mg of weed-ether at 9.5 d of gestation and caesarean section on 21 d of gestation (24 h before the expected date). The trachea and bronchi were stained with Alcianblue alizarin and the organs were examined under a surgical microscope. The results with 11 normal fetal control. Results There was no abnormality in the trachea and bronchus structure in normal control fetus. The number of cartilage tracheal rings in fetal rat with congenital diaphragmatic hernia (CDH) obviously decreased. Of the 7 congenital diaphragmatic hernia mice, 40 had incomplete tracheal soft rings, 12 had varying degrees of tracheal stenosis and were associated with abnormal vascular rings. Fetal rats induced by Nitrofen without congenital diaphragmatic hernia also had a decrease in the number of tracheal rings, with only mild tracheal stenosis. Conclusion Nitrofen induced fetal rat embryos in addition to congenital diaphragmatic hernia with pulmonary dysplasia, sometimes with severe tracheobronchial abnormalities. Tip in embryonic pathogenesis involves a variety of tissue components. Teratogenic agents may interfere with the regulation of the development of tissues and organs.
