目的:探讨电镜冷冻蚀刻技术在Duchenne 型肌营养不良症( DMD) 研究中的应用。方法:电镜观察DMD 及正常对照的骨骼肌和红细胞的冷冻蚀刻样品。结果:DMD 患者红细胞与肌细胞一样,其细胞膜中的蛋白颗粒显著减少。结论:①冷冻蚀刻技术有助于DMD 的诊断,其细胞膜存在缺陷;②可取红细胞代替肌组织进行诊断。
Objective: To investigate the application of electron microscopy cryoetching in the study of Duchenne muscular dystrophy (DMD). Methods: Electron microscopy was used to observe the freeze-dried samples of skeletal muscle and erythrocytes from DMD and normal controls. Results: Erythrocytes in DMD patients, like muscle cells, showed a significant reduction of protein granules in the cell membrane. Conclusion: ① Cryo-etching technique is helpful for the diagnosis of DMD, and its cell membrane is defective; ② Red blood cells can be used instead of muscle tissue for diagnosis.
