目的:探讨附子多糖对小鼠海马糖原合成酶激酶-3β(GSK-3β)蛋白表达的影响。方法:健康雄性C57BL/6J小鼠12只,分为对照组(n=3)和实验组(n=9),实验组分为附子多糖低、中、高3个剂量组,每组各3只,用药剂量分别为50mg/kg、100mg/kg、200mg/kg,对照组腹腔注射等量0.9%氯化钠注射液。6h后断头取脑,剥离海马组织,并用westren blot检测海马神经元中GSK-3β及p-GSK-3β的表达。结果:附子多糖各剂量组GSK-3β表达水平与对照组比较,差异无统计学意义(P>0.05),p-GSK-3β-Ser9/内参蛋白比值高于对照组(P<0.05)。结论:附子多糖影响GSK-3β活性可能是其增加神经细胞发生的作用机制之一。
Objective: To investigate the effect of aconite polysaccharide on the protein expression of glycogen synthase kinase-3β (GSK-3β) in mouse hippocampus. Methods: Twelve healthy male C57BL / 6J mice were divided into control group (n = 3) and experimental group (n = 9). The experimental group consisted of low, medium and high doses of aconite polysaccharide with 3 doses of 3 Only, the dosage was 50mg / kg, 100mg / kg, 200mg / kg, the control group, intraperitoneal injection of 0.9% sodium chloride injection. Six hours later, the brain was decapitated and the hippocampus was dissected. Western blot was used to detect the expression of GSK-3β and p-GSK-3β in hippocampal neurons. Results: Compared with the control group, the expression of GSK-3β in the aconite polysaccharide group had no significant difference (P> 0.05). The ratio of p-GSK-3β-Ser9 / endoprotein was higher than that in the control group (P <0.05). CONCLUSION: The effect of aconite polysaccharide on GSK-3β activity may be one of its mechanisms of increasing neuronal cell.
