研究了罗汉果皂甙提取物对乙醇诱导L-02肝细胞损伤的保护作用。试验结果表明,10~500μmol/L的罗汉果皂甙提取物对培养的L-02细胞无明显毒性;100,200μmol/L的罗汉果皂甙提取物干预组细胞生长状态良好,接近正常组,并显著提高乙醇诱导损伤L-02细胞的存活率(P<0.05)。此外,50~200μmol/L的罗汉果皂甙提取物可显著降低培养上清液天冬氨酸氨基转移酶、乳酸脱氢酶活性及细胞丙二醛水平(P<0.05),明显提高细胞超氧化物歧化酶活性和谷胱甘肽水平(P<0.05)。罗汉果皂甙提取物预处理可保持L-02细胞膜完整性,增强细胞抗氧化性,抑制脂质过氧化反应,对体外乙醇诱导肝细胞损伤有拮抗作用。
The protective effect of extracts of luohan fruit saponins on ethanol induced L-02 hepatocyte injury was studied. The test results showed that 10~500μmol/L locushama saponin extract had no obvious toxicity to cultured L-02 cells; 100,200μmol/L locushama saponin extract intervention group had good cell growth status, close to the normal group, and significantly increased ethanol induction. Survival rate of injured L-02 cells (P<0.05). In addition, 50~200μmol/L extracts of mogrosides significantly reduced aspartate aminotransferase, lactate dehydrogenase activity, and cellular malondialdehyde levels in culture supernatants (P<0.05), and significantly increased cellular superoxide Dismutase activity and glutathione levels (P < 0.05). Pretreatment with extract of Lo Han Guosaponin can maintain the integrity of L-02 cell membrane, enhance cellular antioxidation, inhibit lipid peroxidation, and antagonize ethanol-induced hepatocyte injury in vitro.
