目的:通过检测中度低温下肠上皮细胞屏障氧化应激损伤后炎症因子(TNF-α、IL-1β、IL-6)的表达水平,阐明中度低温对其炎症因子表达的影响及其临床意义。方法:应用Caco-2细胞株建立体外肠上皮细胞屏障模型,分中度低温组、常温组、亚低温组3个组,肠上皮细胞屏障氧化应激损伤后,应用逆转录PCR和实时荧光定量PCR检测细胞内炎症因子mRNA的表达水平,ELISA法检测培养上清中炎症因子的表达水平。结果:肠上皮细胞屏障氧化应激损伤后,中度低温组细胞内炎症因子的mRNA和以及培养上清中炎症因子的质量浓度明显低于常温组和亚低温组。结论:肠上皮细胞屏障氧化应激损伤后,中度低温比常温、亚低温更能抑制炎症因子的分泌,具有重要的临床意义。
OBJECTIVE: To determine the expression of inflammatory cytokines (TNF-α, IL-1β, IL-6) after oxidative stress injury in intestinal epithelial cells at moderate hypothermia, and to clarify the effect of moderate hypothermia on the expression of inflammatory cytokines and its clinical significance significance. METHODS: Caco-2 cell line was used to establish a model of intestinal epithelial cell barrier in vitro. The cells were divided into 3 groups: moderate hypothermia group, normal temperature group and mild hypothermia group. After oxidative stress injury of intestinal epithelial cells, reverse transcription PCR and real- PCR was used to detect the mRNA expression of inflammatory cytokines. The expression of inflammatory cytokines in the culture supernatants was detected by ELISA. Results: After oxidative stress injury of intestinal epithelial cells, the levels of inflammatory cytokines and cytokines in the medium hypothermia group were significantly lower than those in the normal and hypothermia groups. CONCLUSION: After oxidative stress injury of intestinal epithelial barrier, moderate hypothermia can inhibit the secretion of inflammatory cytokines more than normal temperature and mild hypothermia, which has important clinical significance.
