目的探讨Annexin A7在匹罗卡品癫痫小鼠模型脑组织中的表达。方法将30只健康成年雄性C57BL/6小鼠随机分为实验组(n=20)和对照组(n=10),实验组建立癫痫小鼠模型,观察两组行为学特点,分别运用蛋白质免疫印迹和免疫荧光标记技术检测Annexin A7在两组小鼠脑组织中的表达。结果实验组20只小鼠中有18只出现了癫痫持续状态,最终存活8只,其中有6只出现自发性发作,对照组均未出现癫痫持续状态和自发性发作。实验组海马和颞叶皮质中Annexin A7的表达均显著高于对照组(P<0.05);实验组海马和颞叶皮质中Annexin A7表达于神经元,而不表达于星形胶质细胞。结论 Annexin A7可能参与癫痫的发生与发展。
Objective To investigate the expression of Annexin A7 in the brain of pilocarpine-induced epilepsy mice. Methods Thirty healthy adult male C57BL / 6 mice were randomly divided into experimental group (n = 20) and control group (n = 10). Epilepsy mice model was established in the experimental group. The behavioral characteristics of the two groups were observed. Western blotting and immunofluorescence staining were used to detect the expression of Annexin A7 in the brain tissue of both groups. RESULTS: Twenty-eight of 20 mice in the experimental group showed status epilepticus and finally survived in 8 rats. Six of them showed spontaneous seizures, and none of the control group had status epilepticus and spontaneous seizures. The expression of Annexin A7 in hippocampus and temporal cortex of experimental group was significantly higher than that of control group (P <0.05). Annexin A7 in hippocampus and temporal cortex of experimental group was expressed in neurons but not in astrocytes. Conclusion Annexin A7 may be involved in the occurrence and development of epilepsy.
