目的:采用高效液相色谱法建立细圆藤药材的指纹图谱,对多批细圆藤药材进行质量评价,为细圆藤药材质量标准的研究提供一定的科学依据。方法:采用高效液相色谱法,Waters XBridge C18色谱柱(4.6 mm×250 mm,5μm),以乙腈-0.2%磷酸为流动相梯度洗脱,流速1.0 m L·min-1,检测波长327 nm,柱温30℃;建立细圆藤药材HPLC指纹图谱,利用“中药色谱指纹图谱相似度评价系统”2012.1版软件对10批药材进行相似度评价,并利用SPSS 22.0软件对所得10批细圆藤药材色谱数据采用Ward法,平方Euclidean距离度量标准进行聚类分析,并进行主成分分析。结果:以S4号样品的图谱为参照图谱,采用多点校正和Mark峰匹配,10批不同产地的细圆藤药材共标定了11个共有峰,并用对照品指认出绿原酸;10批样品指纹图谱的相似度均>0.9,通过聚类及主成分分析,将10批不同产地的药材分为4类。结论:该方法具有较高的稳定性,方法简便且专属性强,所建立的指纹图谱具有较好的稳定性,对进一步研究细圆藤药效物质基础有重要意义,也为广西壮药细圆藤的质量控制与评价提供了参考。
OBJECTIVE: To establish the fingerprint of Salvia elatiorrhiza with HPLC and to evaluate the quality of Salvia elata.
