目的探讨大蒜素对人直肠癌SW480细胞周期的影响及其与特异性抗癌药物草酸铂联合使用的抗肿瘤作用。方法MTT法检测大蒜素对直肠癌细胞的增殖抑制作用;流式细胞仪检测细胞周期改变;Annexin V-FITC标记法定量检测细胞凋亡;观察大蒜素与草酸铂联合使用后药物对细胞毒活性的改变。结果大蒜素可明显抑制SW480细胞生长,大蒜素作用SW480细胞24 h后,与对照组比较,G0/G1期细胞的百分率明显减少,而G2/M期明显增加(P<0.05),大蒜素与草酸铂联合使用后,草酸铂72 h IC50值由单独应用的9.21μg/ml下降至1.07μg/ml;细胞凋亡率由65.3%增至97%。结论大蒜素可将直肠癌SW480细胞阻滞于G2/M期,大蒜素与草酸铂联合使用可能具有协同抗肿瘤作用。
Objective To investigate the effect of allicin on the cell cycle of human rectal cancer SW480 and the antitumor effect of allicin in combination with the specific antitumor drug oxaliplatin. Methods MTT assay was used to detect the inhibitory effect of allicin on the proliferation of rectal cancer cells. Flow cytometry was used to detect cell cycle changes. Annexin V-FITC method was used to detect the apoptosis of rectal cancer cells. The effects of allicin and oxaliplatin on cytotoxicity Change. Results Allicin significantly inhibited the growth of SW480 cells. Allicin significantly decreased the percentage of cells in G0 / G1 phase and increased G2 / M phase (P <0.05) 24 h after SW480 cells were treated with allicin. Compared with control group, The combination of oxaliplatin and IC50 of oxaliplatin at 72 h decreased from 9.21 μg / ml to 1.07 μg / ml alone, and the apoptosis rate increased from 65.3% to 97%. Conclusion Allicin can block SW480 cells in G2 / M phase. The combination of allicin and oxaliplatin may have a synergistic antitumor effect.
