探讨了大豆及大豆制品中金雀异黄素及大豆黄素对化学致癌物质起始作用的影响。 方法:①小鼠微核试验:对ICR小鼠单剂量腹腔内给予二甲基苯蒽(DMBA),再单剂量经口给予金雀异黄素、大豆黄素。48h后处死小鼠,取外周血及骨髓细胞制作吖啶橙染色标本,计算微核的诱发率。②小鼠多脏器碱性色甘酸二钠(SCG)试验:摘出微核试验小鼠的7种脏器,匀浆后在载玻片上用琼脂糖凝胶包埋、电泳。用溴化乙锭染色后,根据核的泳动观察DNA断裂的程度。 结果与讨论:小鼠微核试验、小鼠多脏器碱性SCG试验均显示金雀异黄素、大
The effects of genistein and daidzein on the initiation of chemical carcinogens in soybean and soybean products were discussed. Methods: 1 Micronucleus test in mice: One dose of dimethyl benzoquinone (DMBA) was intraperitoneally administered to ICR mice, and then genistein and daidzein were administered orally in a single dose. After 48 hours, the mice were sacrificed. Peripheral blood and bone marrow cells were used to prepare acridine orange stained specimens and the induction rate of micronucleus was calculated. 2 Mice multiple organs basic disodium cromoglycate (SCG) test: 7 kinds of organs in micronucleus test mice were extracted, homogenized and embedded in agarose gel on a glass slide and electrophoresed. After staining with ethidium bromide, the degree of DNA fragmentation was observed by the migration of the nucleus.
