目的研究大鼠胶质瘤细胞内生物钟基因Period1(Per1)、Per2mRNA及蛋白的周期表达规律。方法将体外培养的大鼠胶质瘤C6细胞接种于SD大鼠右侧尾状核区,在12h光照和12h黑暗的标准昼夜节律环境下,采用RT-PCR和免疫组织化学方法分别检测成瘤后4、8、12、16、20、24h肿瘤组织中Per1、Per2 mRNA和蛋白的表达。结果在大鼠胶质瘤组织中,Per1、Per2mRNA及蛋白产物呈节律性表达,其表达周期为12h的超日节律。Per1mRNA和蛋白的高表达时间点均位于4、16h,低表达时间点位于12、24h;Per2mRNA和蛋白的高表达时间点位于12、24h,低表达时间点位于8、20h。结论在SD大鼠体内,胶质瘤细胞的Per1、Per2基因呈12h的超日节律表达,提示生物钟基因的表达异常与胶质瘤的发生发展具有相关性。
OBJECTIVE: To study the periodic expression pattern of Perl, Per2 mRNA and protein in rat brain glioma cells. Methods Rat glioma C6 cells cultured in vitro were inoculated into right caudate nucleus caudatum of SD rats. RT-PCR and immunohistochemistry were used to detect tumor formation in 12 h light and 12 h dark day, respectively. Expression of Per1, Per2 mRNA and protein in tumor tissues after 4, 8, 12, 16, 20 and 24 hours. Results The expression of Per1 and Per2 mRNA and protein in rodent glioma tissues was rhythmic, and the expression period was 12 h. Per1 mRNA and protein expression time points were located at 4,16h, low expression time point at 12,24h; Per2mRNA and protein expression time point at 12,24h, low expression time point at 8,20h. Conclusion Peri and Per2 genes of glioma cells were up-regulated in 12-day rhythms in SD rats, suggesting that the abnormal expression of circadian clock genes is correlated with the occurrence and development of gliomas.
