目的:观察全蚕粉对2型糖尿病胰岛素受体信号传导通路的影响,探讨全蚕粉逆转2型糖尿病胰岛素抵抗的分子作用机理。方法:以高热量饲料喂养大鼠8周加小剂量链脲佐菌素(STZ,25mg/kg)腹腔注射建立2型糖尿病大鼠模型。应用全蚕粉进行干预,以阿卡波糖片作为阳性对照药,用Real-time PCR和Western blotting技术,观察全蚕粉对Akt,InsR,PI3K基因及其蛋白表达的影响。结果:与模型组比较,全蚕粉组0.5、1.0、2.0g/kg肝组织Akt,InsR,PI3K mRNA的相对表达量无显著性差异,肝组织Akt,InsR,PI3K蛋白表达量显著上调(P﹤0.05)。结论:全蚕粉对2型糖尿病大鼠的影响可能在翻译水平上通过PI3K信号传导途径增强胰岛素生物效应,从而逆转2型糖尿病大鼠胰岛素抵抗。
OBJECTIVE: To investigate the effect of silkworm powder on insulin receptor signaling pathway in type 2 diabetes mellitus (T2DM) and to explore the molecular mechanisms of silkworm polysaccharide in reversing insulin resistance in type 2 diabetes mellitus (T2DM). Methods: Type 2 diabetic rats were established by intraperitoneal injection of high-calorie diet for 8 weeks and low-dose streptozotocin (STZ, 25 mg / kg). The whole silkworm powder was used for intervention. Acarbose was used as a positive control drug. Real-time PCR and Western blotting were used to observe the effect of silkworm powder on the expression of Akt, InsR, PI3K and their proteins. Results: Compared with the model group, there was no significant difference in the expression of Akt, InsR and PI3K mRNA in the liver tissues of 0.5, 1.0 and 2.0 g / kg of whole silkworm powder group, while the expression of Akt, InsR and PI3K in liver tissues was significantly increased (P <0.05). Conclusion: The effect of whole silkworm powder on type 2 diabetic rats may enhance the insulin biological effects through the PI3K signaling pathway at the translational level, thus reversing the insulin resistance in type 2 diabetic rats.
