目的:探讨尖端电喷雾萃取电离质谱技术(tipspray ionization mass spectrometry,Tip-sprayMS)对食管癌(esophageal cancer,EC)诊断的应用.方法:收集22例EC及配对的癌旁组织,应用Tip-spray-MS技术对组织样品进行快速分析,调节Tip-spray离子源为正离子模式,质量范围50-1000 Da,喷雾电压为3.5 kV,喷雾距离为2 cm,透镜电压为65.0 V,离子传输管温度为150℃.运用Matlab软件主成分分析(principal component analysis,PCA)方法对所获质谱指纹谱进行统计学分析.结果:EC与癌旁组织的质谱指纹谱图中相对丰度有明显差异的质荷比(m/z)为104、154、175、611、655、699、743、787、831等处,其中m/z为611、655、699处丰度差异最显著,其所对应的物质为卟啉,运用PCA方法能够良好地区分EC及癌旁组织.结论:Tip-spray-MS联合PCA能有效鉴别EC及癌旁正常组织,有助于EC的早期、快速诊断.

Statistical analysis of MS fingerprinting was performed using the principal component analysis (PCA) method of Matlab software.Results: There was a significant difference in the relative abundance between EC and adjacent tissues (M / z) was 104,154,175,611,655,699,743,787,831, etc. Among them, m / z was the most significant difference at the abundance of 611,655,699, and its corresponding substance Porphyrin and PCA could be used to distinguish EC from adjacent tissues.Conclusion Tip-spray-MS combined with PCA can effectively differentiate EC from adjacent normal tissues and contribute to the early and rapid diagnosis of EC.
