目的 在高脂饮食诱导胰岛素抵抗的基础上 ,观察饮食对大鼠肝脏中蛋白酪氨酸磷酸酶 1B(PTP1B)表达的影响。方法 选取雄性Wistar大鼠 3 0只 ,随机分为正常对照组 10只 ,给予基础饲料 ;模型组 2 0只 ,给予高脂饲料。模型组大鼠给予高脂喂养 4周后 ,随机分为 2亚组 :胰岛抵抗组 ,继续高脂饮食 ;饮食组 ,给予低脂饮食干预。干预 6周后 ,用蛋白印迹法检测各组大鼠肝组织中PTP1B蛋白含量。结果 在胰岛素抵抗状态时 ,大鼠肝组织中PTP1B蛋白与对照组相比 ,含量增加 98 3 % (t =9 3 3 5 ,P <0 0 0 1) ;饮食干预能明显降低PTP1B的表达 ,与胰岛素抵抗组相比减少 3 2 7% (t =4 0 76,P <0 0 0 1)。结论 饮食干预能改善机体胰岛素抵抗 ,可能与降低肝脏组织中PTP1B蛋白表达有关。
Objective To observe the effect of diet on the protein tyrosine phosphatase 1B (PTP1B) expression in rat liver on the basis of insulin resistance induced by high-fat diet. Methods Thirty male Wistar rats were randomly divided into normal control group (n = 10), basal diet and model group (n = 20). Rats in model group were fed with high-fat diet for 4 weeks and were randomly divided into 2 subgroups: Islet-resistant group, and continued high-fat diet; the diet group was given low-fat diet intervention. Six weeks after the intervention, the content of PTP1B protein in the liver tissue of each group was detected by Western blotting. Results Compared with the control group, PTP1B protein in liver tissue of rats increased by 98.3% (t = 93.335, P <0.01), and the diet intervention could significantly reduce the expression of PTP1B, Compared with the insulin resistance group, a decrease of 37.2% (t = 4076, P <0.01). Conclusion Diet intervention can improve the body’s insulin resistance, which may be related to the decrease of PTP1B protein expression in the liver.
