目的:探讨大鼠心肌缺血再灌注后不同时间点心脏和肝脏中缺氧诱导因子-1(hypoxia-inducible factor 1,HIF-1)及血管内皮生长因子(vascular endothelial growth factor,VEGF)的变化。方法:结扎雄性成年SD大鼠冠状动脉左前降支,缺血30min,再灌注6 h(I30minR6h)或24 h(I30minR24h),real-time PCR法检测心肌组织中HIF-1α和VEGF的mRNA水平,Western blot法检测心肌以及肝脏组织中HIF-1α、HIF-1β和VEGF蛋白表达的改变。结果:与假手术组相比,I30minR6h组心肌中HIF-1α和VEGFmRNA和蛋白水平明显增加,但I30minR24h组无明显变化。心肌组织中HIF-1β蛋白水平及肝脏中HIF-1α、HIF-1β和VEGF蛋白水平在不同再灌注时间点无明显改变。结论:心肌缺血再灌注影响心肌HIF-1和VEGF的表达,但对肝脏中HIF-1和VEGF的表达无影响,且其对心肌HIF-1和VEGF表达的影响与再灌注时间相关。
Objective: To investigate the changes of hypoxia-inducible factor 1 (HIF-1) and vascular endothelial growth factor (VEGF) in the heart and liver at different time points after myocardial ischemia-reperfusion in rats . Methods: The left anterior descending coronary artery of adult male Sprague Dawley rats was ligated for 30 min, reperfusion at 6 h (I30 min R6 h) or 24 h (I30 min R24 h), and the mRNA expression of HIF-1α and VEGF in myocardium was detected by real- The changes of HIF-1α, HIF-1βand VEGF protein expression in myocardium and liver were detected by Western blot. Results: Compared with the sham operation group, the levels of HIF-1α and VEGF mRNA and protein in the myocardium of I30minR6h group increased significantly, but there was no significant change in I30minR24h group. The level of HIF-1|Â protein in myocardium and the level of HIF-1|Á, HIF-1|Â and VEGF in liver did not change significantly at different reperfusion time points. CONCLUSION: Myocardial ischemia-reperfusion affects the expression of HIF-1 and VEGF, but has no effect on the expression of HIF-1 and VEGF in the liver. The effect of HIF-1 and VEGF on the expression of HIF-1 and VEGF is related to the reperfusion time.
