目的观察加味良附丸联合5-Fu对人胃癌细胞SGC-7901裸鼠移植瘤生长抑制作用及其对肿瘤组织中核转录因子-kappaBp65(NF-ΚBp65)表达的影响。方法建立人胃癌细胞SGC-7901移植瘤动物模型,随机分为模型组、5-FU组及加味良附丸大、中、小剂量组、加味良附中剂量+5-FU组(联合给药组)。给药10 d,计算肿瘤抑制率,免疫组化检测肿瘤组织中NF-ΚBp65的表达。结果联合给药组、5-FU组、加味良附丸大、中、小剂量组抑瘤率分别为54%、45%、31%、28%、17%,联合给药组与加味良附丸大、中、小剂量比,差异显著有统计学意义(P<0.05);模型组肿瘤组织中NF-ΚBp65表达明显高于各给药组,差异显著有统计学意义(P<0.05);联合给药组肿瘤组织中NF-ΚBp65表达低于加味良附丸大、中、小剂量组,差异明显有统计学意义(P<0.05)。结论加味良附丸联合5FU组抑瘤率高于单纯加味良附丸高、中、低剂量组及5-FU组,联合给药抑制NF-ΚBp65表达方面优于加味良附丸高剂量组及5-Fu组,提示中西药联合具有协同作用。
Objective To observe the effects of Jiaweilianpill combined with 5-Fu on the growth of human gastric cancer SGC-7901 xenografted in nude mice and its effect on the expression of nuclear factor-kappaBp65 (NF-κBp65) in tumor tissues. Methods Animal models of SGC-7901 xenografts in human gastric cancer cells were established and randomly divided into model group, 5-FU group and Jiaweilianfu pill large, medium and small dose groups, Jiaweilianzhong medium dose + 5-FU group (combination group) . After administration for 10 days, the tumor inhibition rate was calculated and the expression of NF-κBp65 in tumor tissue was detected by immunohistochemistry. Results The tumor inhibition rates of the combination group, 5-FU group and Jiaweilianfu pill groups were 54%, 45%, 31%, 28% and 17% (P <0.05). The expression of NF-κBp65 in the model group was significantly higher than that in the other groups (P <0.05). The combination of The expression of NF-ΚBp65 in the tumor tissue of the drug group was lower than that of the Jiawei lianfu pill group, and the difference was statistically significant (P <0.05). Conclusions The antitumor rate of Jiaweiliangfu pills combined with 5FU group is higher than that of Jiaweiliangfu pill high, medium and low dose groups and 5-FU group. The combination of Jiaweiliangfu pills and 5FU group is superior to Jiaweilifu pill high-dose group in inhibiting the expression of NF-κBp65 5-Fu group, suggesting that the combination of Chinese and Western medicine has a synergistic effect.
