目的:观察猫爪草总皂苷体外对人非小细胞肺癌A549细胞活性的影响,初步探讨其可能的作用机制。方法:采用系统溶剂法提取猫爪草总皂苷,3H-TdR掺入法和集落形成实验观察猫爪草总皂苷对A549细胞增殖的影响;流式细胞仪检测其对A549细胞凋亡和周期的影响。结果:猫爪草总皂苷对A549细胞增殖和集落形成均有较好的抑制作用,呈现较好的量效关系,可促进A549细胞的早期凋亡,细胞周期出现G0/G1期的阻滞。结论:猫爪草总皂苷能较好的抑制A549细胞增殖,其机制可能与诱导凋亡、G0/G1期阻滞有关。
Objective: To observe the effect of Radix Ranunculus total saponins on human non-small cell lung cancer cell line A549 in vitro and to explore its possible mechanism. Methods: The systemic solvent method was used to extract the total saponin of Radix Ranunculus. 3H-TdR incorporation and colony formation assay were used to observe the effect of Radix Ranunculus total saponin on the proliferation of A549 cells. Flow cytometry was used to detect the apoptosis and cell cycle of A549 cells influences. Results: The total saponin of cat claw grass had a good inhibitory effect on the proliferation and colony formation of A549 cells, showing a good dose-effect relationship, which could promote the early apoptosis of A549 cells and the arrest of G0 / G1 phase in the cell cycle. Conclusion: The total saponin of cat claw grass can better inhibit the proliferation of A549 cells, which may be related to the induction of apoptosis and the arrest of G0 / G1 phase.
