目的观察不同浓度的碱性成纤维细胞生长因子(bFGF)对体外培养中划痕损伤后ECV304细胞迁移的影响。方法在体外细胞划痕损伤模型中应用显微电视电脑图像处理系统定量测定不同浓度(0、5、10、15μg/L)bFGF引起的ECV304细胞迁移的变化,用光镜与扫描电镜观察bFGF引起的迁移细胞的形态变化。结果与不加bFGF的对照组比较,低浓度(5μg/L)时bFGF对ECV304细胞的迁移呈促进作用;高浓度时(15μg/L)呈抑制作用。迁移细胞表面有众多丝状伪足。结论bFGF对体外培养的ECV304细胞的迁移有双相调节作用,低浓度时(5μg/L)促进细胞迁移,高浓度时(15μg/L)抑制细胞迁移。迁移细胞表面伪足丰富,以丝状伪足为主。
Objective To observe the effects of different concentrations of basic fibroblast growth factor (bFGF) on the migration of ECV304 cells induced by scratches in vitro. Methods The changes of migration of ECV304 cells induced by bFGF at different concentrations (0, 5, 10, and 15 μg / L) were determined by microdisplay computerized image processing system in vitro. The changes of bFGF induced by bFGF were observed with light microscope and scanning electron microscope The morphological changes of migrating cells. Results Compared with the control group without bFGF, bFGF promoted the migration of ECV304 cells at low concentration (5μg / L), and inhibited the proliferation of ECV304 cells at high concentration (15μg / L). There are numerous filopodia on the surface of migrating cells. Conclusion bFGF can regulate the migration of ECV304 cells in vitro. When the concentration of bFGF is 5μg / L, the migration of ECV304 cells is inhibited. At high concentration, bFGF inhibits the migration of ECV304 cells. Migratory cell surface pseudopodia rich in filopodia-based.
