目的:通过对子宫内膜癌组织基因表达谱的层次聚类分析,寻找子宫内膜癌“高危”患者。方法:以32例内膜样子宫内膜腺癌为实验组,36例正常子宫内膜组织混合作为对照组。分别用Cy3和Cy5标记实验组和对照组。用含有13 824个基因的人全基因表达谱cDNA芯片分析32例内膜癌的基因表达谱,筛选差异表达基因,并对基因表达谱进行层次聚类分析。结果:按肿瘤的分化程度、分期以及分化分期相结合进行分组,分别筛选出差异表达基因46、12和138个(P<0.001)。对32例内膜癌进行层次聚类分析的结果与肿瘤的分化、FIGO分期分类基本相符,并且能够发现一些子宫内膜癌“高危”患者。结论:对子宫内膜癌基因表达谱的研究可以发现与子宫内膜癌浸润、转移相关的基因;基因表达谱的聚类分析可以在术前帮助判断子宫内膜癌的“高危”病患。
OBJECTIVE: To search for “high risk” endometrial cancer patients by hierarchical cluster analysis of endometrial cancer gene expression profiles. Methods: 32 cases of endometrial adenocarcinoma as experimental group and 36 cases of normal endometrial tissue as control group. The experimental group and the control group were labeled with Cy3 and Cy5 respectively. Gene expression profiles of 32 cases of endometrial carcinoma were analyzed by cDNA microarray containing 13 824 genes. The differentially expressed genes were screened and the cluster analysis of gene expression profiles was performed. Results: According to the degree of tumor differentiation, staging and differentiation staging, 46,12,12 differentially expressed genes were screened (P <0.001). Hierarchical cluster analysis of 32 cases of endometrial cancer results and tumor differentiation, FIGO staging classification basically consistent, and can be found in some endometrial cancer “high risk” patients. Conclusion: The gene expression profile of endometrial cancer can be found in the endometrial cancer invasion and metastasis-related genes; cluster analysis of gene expression profiling can help determine the “high risk” of endometrial cancer patients before surgery.
