本文用H_2O_2对正常人、阵发性睡眠性血红蛋白尿症(PNH)、再生障碍性贫血(再障)及其他贫血病人红细胞进行处理,然后测定其脂质过氧化产物丙二醛(MDA)含量。PNH红细胞MDA含量明显高于正常人、再障及其他贫血病人(P<0.01)。经H_2O_2处理后红细胞膜SDS-PAGE图谱显示区带1、2、3、5均减少;膜巯基含量随H_2O_2浓度加大或作用时间延长而逐渐减少。测定红细胞脂质过氧化方法简单、敏感,可协助临床对PNH、再障及其他贫血进行鉴别诊断。过氧化与溶血关系很密切,也可以从此领域探讨PNH溶血的机理。
H 2 O 2 was used to treat erythrocytes in normal subjects, paroxysmal nocturnal hemoglobinuria (PNH), aplastic anemia (aplastic anemia) and other anemia patients, and then the content of malondialdehyde (MDA) . PNH erythrocyte MDA content was significantly higher than normal, aplastic anemia and other anemia patients (P <0.01). The H 2 O 2 treated erythrocyte membrane SDS-PAGE pattern showed the band 1,2,3,5 were reduced; membrane sulfhydryl content with the H 2 O 2 concentration increases or the role of prolonged and gradually decreased. Determination of erythrocyte lipid peroxidation method is simple and sensitive, can help clinical differential diagnosis of PNH, aplastic anemia and other anemia. Peroxidation and hemolysis are closely related, but also from this area to explore the mechanism of PNH hemolysis.
