目的:探讨补肾方药左归丸与右归丸含药鼠血清对皮质酮(CORT)损伤型大鼠海马病理细胞模型形态结构的影响。方法:以新生SD乳鼠的海马组织为材料,原代培养海马神经细胞;以10~(-4)mol/LCORT处理培养的细胞制作大鼠海马病理细胞模型,并用RU 38486进行拮抗。采用NSE和NissI染色、FDA/PI、SYTO-13/PI双重染色法等观察模型细胞形态结构变化及左归丸与右归丸含药鼠血清的保护作用。结果:与正常对照组比较,加入10~(-4)mol/L CORT以处理培养的细胞,可使海马神经细胞明显受损,死细胞增多;左归丸与右归丸含药鼠血清均可使蓝色荧光的活细胞显著增多,呈红色荧光的死亡细胞明显减少。结论:加入10~(-4)mol/L CORT处理神经细胞可以制作海马病理细胞模型,补肾方药左归丸或右归丸含药血清对CORT损伤型海马病理细胞模型有明显的保护作用。
Objective: To investigate the effect of Zuogui Pill and Yougui Pill containing drug serum on the morphological and ultrastructure of hippocampal pathological model rats with corticosterone (CORT) -induced injury. Methods: Hippocampal neurons were cultured in neonatal SD suckling rats. Primary cultured rat hippocampal neurons were treated with 10 ~ (-4) mol / L LCORT and the cells were treated with RU 38486. NSE and NissI staining, FDA / PI, SYTO-13 / PI double staining and other observations of the morphological changes of the model cells and Zuogui pill and Youguiwan containing rat serum protective effect. Results: Compared with the normal control group, the cultured cells were treated with 10 ~ (-4) mol / L CORT, which significantly damaged the hippocampal neurons and increased the number of dead cells. Serum Zuogui Pill and Yougui Wan containing drugs Can make the blue fluorescence of living cells increased significantly, the red fluorescent death cells was significantly reduced. CONCLUSION: The cultured hippocampal neurons with 10 ~ (-4) mol / L CORT can make the pathological cell model of hippocampus. The Bushen Prescriptions Zuogui Pill or Yougui Pill containing serum have significant protective effect on CORT - injured hippocampal CAPS.
