为研究不同鸡种骨骼肌卫星细胞生肌决定因子5(Myf5)基因表达差异,试验通过提取大围山微型鸡和艾维茵肉鸡骨骼肌卫星细胞并进行两个鸡种骨骼肌卫星细胞增殖分化差异及Myf5基因表达差异研究。结果显示:两个鸡种骨骼肌卫星细胞生长曲线呈“S”形,大围山微型鸡骨骼肌卫星细胞生长速度较低;在分化过程中,艾维茵肉鸡骨骼肌卫星细胞分化能力高于大围山微型鸡,且艾维茵肉鸡骨骼肌卫星细胞的Myf5 mRNA表达量显著高于大围山微型鸡(P<0.05)。本试验表明:Myf5基因是影响家禽肌肉生长发育的重要候选基因,且细胞体外培养对家禽分子育种研究具有一定科学指导意义。
In order to study the difference of Myf5 gene expression in skeletal muscle satellite cells of different chicken breeds, the skeletal muscle satellite cells of Dawei Mountain mini-broiler and Avian broiler were extracted and the proliferation and differentiation of two chicken skeletal muscle satellite cells Differences and Myf5 gene expression differences. The results showed that the growth curves of skeletal muscle satellite cells of two chicken breeds were “S” shaped, and the growth of satellite cells in Dawei Mountain miniature chicken skeletal muscle was relatively slow. During the differentiation process, the skeletal muscle satellite cells differentiation ability (P <0.05). The Myf5 mRNA expression in skeletal muscle satellite cells of Weiwei broiler chicken was significantly higher than that of Dawei Mountain miniature chicken (P <0.05). This experiment shows that: Myf5 gene is an important candidate gene affecting poultry muscle growth and development, and cell culture in vitro molecular breeding of poultry has a certain scientific significance.
