目的观察乳清酸左卡尼汀对胃溃疡的治疗作用,并探讨其抗溃疡的相关机制。方法大鼠胃溃疡造模,应用HE染色观察大鼠胃溃疡创面的愈合程度,Elisa法测胃溃疡组织IL-8,IL-10的表达情况;应用免疫组化法测胃溃疡组织EGF的表达情况。结果乳清酸左卡尼汀低剂量组25 mg/(kg.d)、乳清酸左卡尼汀中剂量组50 mg/(kg.d)、乳清酸左卡尼汀高剂量组100 mg/(kg.d)对胃溃疡均有治疗作用,中剂量效果接近于奥美拉唑。乳清酸左卡尼汀能够降低组织MDA含量,增强SOD活性,通过抗氧化应激治疗溃疡。乳清酸左卡尼汀通过抑制胃溃疡组织炎症因子IL-8,10表达而起到抗氧化作用。乳清酸左卡尼汀的抗溃疡作用与其促进表皮生长因子(EGF)表达有关。结论乳清酸左卡尼汀对胃溃疡有治疗作用,其机制可能与抗氧化应激和促进EGF表达有关。
Objective To observe the therapeutic effect of L-carnitine on gastric ulcer and to explore its mechanism of anti-ulcer. Methods The model of gastric ulcer was established in rats. HE staining was used to observe the wound healing of gastric ulcer in rats. Elisa method was used to detect the expression of IL-8 and IL-10 in gastric ulcer tissue. The expression of EGF was detected by immunohistochemistry Happening. Results L-carnitine low dose group 25 mg / (kg.d), Lactic acid L-carnitine medium dose group 50 mg / (kg.d), Lactic acid L-carnitine high dose group 100 mg / (kg.d) have a therapeutic effect on gastric ulcer, the effect of medium dose close to omeprazole. Lactic acid levocarnitine can reduce tissue MDA content, increase SOD activity, anti-oxidant stress treatment of ulcers. Lactic acid levocarnitine inhibits the gastric ulcer tissue inflammation factor IL-8,10 expression and play an antioxidant role. The anti-ulcer effect of L-carnitine is related to the promotion of epidermal growth factor (EGF) expression. Conclusion Lactic acid levocarnitine has a therapeutic effect on gastric ulcer. The mechanism may be related to the anti-oxidative stress and EGF expression.
