目的建立辅酶A有关物质测定的HPLC方法。方法采用C18柱(200mm×4.6mm,5μm),以磷酸盐缓冲溶液(pH6.5)-甲醇(90∶10)为流动相,流速:1.0ml/min,检测波长:259nm,柱温:25℃,对辅酶A中的有关物质进行测定。结果主成分峰与相邻杂质峰的分离度均大于1.5。结论该方法能够很好的分离辅酶A中的有关物质,并对其进行控制。
Objective To establish a HPLC method for the determination of coenzyme A related substances. Methods The mobile phase consisted of C18 column (200 mm × 4.6 mm, 5 μm), phosphate buffer (pH 6.5) and methanol (90:10), the flow rate was 1.0 ml / min, the detection wavelength was 259 nm and the column temperature was 25 ℃, the coenzyme A in the determination of related substances. As a result, the resolution of the main component peak and the adjacent impurity peak were both greater than 1.5. Conclusion This method can be a good separation of coenzyme A in the relevant substances, and its control.
