目的探讨内皮素(ET-1),基质金属蛋白酶-1(MMP-1)在口腔癌变发病机制中的作用和意义。方法采用免疫组化染色SABC法对人口腔粘膜白斑48例、白斑癌变患者16例及正常口腔粘膜16例的粘膜鳞状上皮细胞ET-1表达进行定量性分析。结果MMP-1在正常口腔粘膜,粘膜白斑,白斑癌变中表达率分别为25%,72.92%,75%,3组间χ2检验,正常组与白斑组,白斑癌变组均有显著性差异,P<0.05,白斑组与白斑癌变组无显著性差异,P>0.05。ET-1在正常口腔粘膜,粘膜白斑,白斑癌变中表达率分别为6.25%,79.17%,87.5%。3组间χ2检验,正常组与白斑组,白斑癌变组均有显著性差异,P<0.05,白斑组与白斑癌变组无显著性差异,P>0.05。结论MMP-1和ET-1的阳性表达对于白斑的诊断有重要意义,但不能作为白斑癌变的诊断指标。
Objective To investigate the role and significance of endothelin (ET-1) and matrix metalloproteinase-1 (MMP-1) in the pathogenesis of oral carcinogenesis. Methods Immunohistochemical SABC method was used to quantitatively analyze ET-1 expression in mucosal squamous epithelium of 48 cases of oral leukoplakia, 16 cases of leukoplakia and 16 cases of normal oral mucosa. Results The expression rates of MMP-1 in normal oral mucosa, leukoplakia and leukoplakia were 25%, 72.92% and 75%, respectively. There was significant difference between normal group, leukoplakia group and leukoplakia group <0.05, no significant difference between leukoplakia group and leukoplakia group (P> 0.05). The expression rates of ET-1 in normal oral mucosa, leukoplakia and leukoplakia were 6.25%, 79.17% and 87.5% respectively. There was significant difference between normal group, leukoplakia group and leukoplakia group (P <0.05). There was no significant difference between leukoplakia group and leukoplakia group (P> 0.05). Conclusion The positive expression of MMP-1 and ET-1 is of great significance for the diagnosis of leukoplakia, but not for the diagnosis of leukoplakia.
