目的 研究胃癌组织内血管内表皮生长因子 (vascularendothelialgrowthfactor ,VEGF)及其受体Flt、KDRmRNA的表达及与胃癌生物学行为的相关性。方法 应用半定量逆转录聚合酶链反应(RT PCR)法检测了 6 0例手术切除、病理证实的胃癌组织内的VEGF及受体Flt、KDRmRNA。结果 VEGF、Flt、KDRmRNA在胃癌组织内的阳性表达率分别为 73.3%、86 .7%和 80 .0 % ,明显高于癌旁组织 43.3%、33.0 %和 30 .0 % (P <0 .0 1) ;伴有淋巴结转移组胃癌的VEGF、Flt、KDR的阳性表达率为90 .3%、80 .6 %和 96 .8% ,明显高于无淋巴结转移组 5 1.7%、41.3%和 5 5 .1% (P <0 .0 1) ;VEGF、KDR在高分化型胃癌组阳性表达为 40 .0 %、46 .7% ,明显低于低分化、未分化型胃癌组 ,Flt在高分化型胃癌组其阳性表达为 73 .3 % ,低分化、未分化型胃癌组阳性表达为 87.5 % (P >0 .0 5 ) ,VEGF、Flt、KDRmRNA阳性表达与胃癌组织浸润深度无关。结论 VEGF、Flt、KDR与胃癌的一些生物学行为有一定的相关性 ,VEGF、Flt、KDR的高表达有助于胃癌的预后评估
Objective To study the expression of vascular endothelial growth factor (VEGF) and its receptors, Flt and KDR mRNA in gastric cancer tissues and its correlation with biological behavior of gastric cancer. Methods Semiquantitative reverse transcription polymerase chain reaction (RT PCR) was used to detect VEGF, receptor Flt, and KDR mRNA in 60 specimens of surgically-removed and pathologically confirmed gastric cancer. Results The positive expression rates of VEGF, Flt and KDR mRNA in gastric cancer tissues were 73.3%, 86.7% and 80.0 %, respectively, which were significantly higher than those in adjacent tissues (43.3%, 33.0 % and 30.0%) (P < .0. 0 1) The positive expression rates of VEGF, Flt, and KDR in gastric cancer patients with lymph node metastasis were 90.3%, 80.6 %, and 96.8%, which were significantly higher than those in patients without lymphatic metastasis 5.7%, 41.3%, and 5.7%. The positive expression of VEGF and KDR in the highly differentiated gastric cancer group was 40.0 % and 46.7%, which was significantly lower than that in the poorly differentiated and undifferentiated gastric cancer group. Flt The positive expression of highly differentiated gastric cancer group was 73. 3%, and the positive expression of poorly differentiated and undifferentiated gastric cancer group was 87.5% (P > 0.05). The positive expression of VEGF, Flt and KDR mRNA was not related to the depth of invasion of gastric cancer tissue. Conclusion VEGF, Flt and KDR have some correlation with some biological behaviors of gastric cancer. The high expression of VEGF, Flt and KDR contributes to the prognosis of gastric cancer.
