目的探讨丹酚酸B对慢性血清病肾炎大鼠模型的影响及肾脏保护作用。方法通过改良后的免疫学方法制备慢性血清病肾炎大鼠模型,造模成功后给予丹酚酸B(100,50 mg·kg~(-1))连续灌胃5周,探讨丹酚酸B对慢性血清病肾炎大鼠肾脏指数、24h尿蛋白、血清肌酐(Scr)、尿素氮(BUN)、白蛋白(ALB)、总蛋白(TP)、总胆固醇(TC)、甘油三酯(TG)、丙二醛(MDA)、超氧化物歧化酶(SOD)、白细胞介素1α(IL-1α)、白细胞介素10(IL-10)含量的影响。结果与模型组比较,丹酚酸B高、低剂量组可以降低慢性血清病肾炎大鼠肾脏指数(P<0.01);降低24 h尿蛋白排泄量(P<0.05);降低Scr、BUN的含量(P<0.05,P<0.01);降低血清中TC、TG(P<0.05,P<0.01)的含量,升高血清中ALB、TP的含量(P<0.05,P<0.01);降低血清中MDA的含量(P<0.05,P<0.01),升高血清中SOD的含量(P<0.05,P<0.01);降低血清中IL-1α、IL-10的含量(P<0.05,P<0.01)。结论丹酚酸B可以改善慢性血清病肾炎大鼠肾脏功能,调节血脂异常状态,其机制可能与抗脂质过氧化及调节细胞因子平衡有关。
Objective To investigate the effect of salvianolic acid B on chronic serum nephritis rat model and its protective effect on kidney. Methods A rat model of chronic serum nephritis was established by modified immunological method. After the model was established, salvianolic acid B (100, 50 mg · kg -1) was administered continuously for 5 weeks to investigate the effects of salvianolic acid B The renal index, 24h urinary protein, serum creatinine (Scr), blood urea nitrogen (BUN), albumin (ALB), total protein (TP), total cholesterol (TC), triglyceride (TG) , Malondialdehyde (MDA), superoxide dismutase (SOD), interleukin-1α (IL-1α) and interleukin-10 (IL-10) Results Compared with the model group, the salvianolic acid B high and low dose groups could reduce the renal index of chronic serum nephritis rats (P <0.01), reduce the urinary protein excretion 24 h (P <0.05), and decrease the content of Scr and BUN (P <0.05, P <0.01), decrease the level of serum TC, TG (P <0.05, P <0.01) and increase the content of ALB and TP in serum (P <0.05, P <0.01) (P <0.05, P <0.01), increase the content of SOD in serum (P <0.05, P <0.01), and decrease the contents of IL-1α and IL-10 in serum ). Conclusion Salvianolic acid B can improve renal function and regulate dyslipidemia in chronic serum nephritis rats. The mechanism may be related to anti-lipid peroxidation and regulation of cytokine balance.
