人参皂甙Re对大鼠急性缺血再灌注心肌细胞凋亡及相关基因蛋白表达的影响(英文)

背景:心肌细胞缺血再灌注损伤近来已成为临床研究的热点,但人参及其提取物对急性缺血再灌注损伤后心肌细胞凋亡的作用尚未阐明。目的:观察人参皂甙Re对急性缺血再灌注心肌细胞凋亡及Bcl-2,Bax,Bad和Fas基因蛋白表达的影响,探讨人参皂甙Re抑制心肌细胞凋亡的可能机制。设计:随机对照实验研究。地点、对象和干预:本实验在湖北省武汉市同济医院动物实验中心完成。实验共用Wistar大鼠15只,雌雄不分。按随机数字表法分为假手术组、缺血再灌注组、人参皂甙Re治疗组,每组各5只。建立Wistar鼠缺血再灌注心脏模型。应用原位末端标记法检测心肌凋亡细胞,并进行细胞计数;原位杂交和免疫组化检测Bcl-2mRNA和基因表达产物的蛋白质,通过图像分析系统测量阳性染色区域的平均吸光度值进行量化分析,以观察人参对大鼠心肌缺血再灌注心肌细胞凋亡的影响及基因表达。主要观察指标:人参皂甙Re治疗与否对急性大鼠缺血再灌注心肌细胞凋亡及Bcl-2,Bax,Bad和Fas基因蛋白表达的影响。结果:①假手术组未发现心肌凋亡细胞。缺血再灌注组心肌凋亡细胞数为(134±46)个/视野,人参皂甙Re治疗组为(91±19)个/视野,两组间差异有显著性意义(t=4.63,P<0.01)。②免疫组织化学检测发现缺血再灌注组及人参皂甙Re治疗组Bcl-2,Bax,Bad和Fas基因蛋? BACKGROUND: Myocardial ischemia-reperfusion injury has recently become a hot spot in clinical research. However, the effect of ginseng and its extracts on myocardial cell apoptosis after acute ischemia-reperfusion injury has not been elucidated. Objective: To observe the effect of ginsenoside Re on cardiomyocyte apoptosis and the expression of Bcl-2, Bax, Bad and Fas gene proteins in acute ischemia-reperfusion injury, and to explore the possible mechanism of ginsenoside Re inhibiting cardiomyocyte apoptosis. Design: Randomized controlled experimental study. Location, subject and intervention: This experiment was performed at the Animal Experimental Center of Tongji Hospital, Wuhan, Hubei Province. 15 Wistar rats were used in the experiment, regardless of sex. According to the random number table method, they were divided into sham operation group, ischemia reperfusion group, and ginsenoside Re treatment group, 5 in each group. Wistar rat ischemic reperfusion cardiac model was established. Apoptotic myocytes were detected by in situ end-labeling and cell counts were performed. Bcl-2 mRNA and gene expression products were detected by in situ hybridization and immunohistochemistry. The average absorbance values ​​of the positively stained areas were measured by an image analysis system for quantitative analysis. To observe the effects of ginseng on myocardiac apoptosis induced by myocardial ischemia and reperfusion in rats and gene expression. MAIN OUTCOME MEASURES: Effects of treatment with ginsenoside Re on cardiomyocyte apoptosis and expression of Bcl-2, Bax, Bad, and Fas genes in rats with acute ischemia-reperfusion. RESULTS: 1No apoptotic cells were found in the sham operation group. The number of apoptotic cells in the ischemia-reperfusion group was (134±46)/field, and the ginsenoside Re group was (91±19)/field. There was a significant difference between the two groups (t=4.63, P< 0.01). 2 Immunohistochemical detection of Bcl-2, Bax, Bad and Fas genes in ischemia-reperfusion group and ginsenoside Re treatment group.
