目的通过酸和胆汁混合灌注制备家兔混合反流性食管炎动物模型。方法采用盐酸和猪胆汁混合灌注建立家兔混合反流性食管炎模型,模型组以灌注不同时间分为4 d组、7 d组及10 d组,正常组则灌注生理盐水;分别在肉眼下、光镜及电镜下观察各组食管变化,并进行分级比较。结果混合灌注后食管下端病理改变明显。3组模型家兔食管下段存在不同程度黏膜粗糙、充血、黏膜下血管增生,见散在或广泛糜烂,并可见不同程度溃疡。3组模型家兔食管肉眼积分差异有统计学意义(P<0.01)。模型家兔可见部分鳞状上皮固缩脱落,固有层嗜酸性细胞、中性粒细胞浸润,黏膜下层内血管扩张、充血,见不同程度的溃疡、组织坏死,食管肌层也可见不同程度炎性细胞浸润。3组模型家兔食管病理积分差异有统计学意义(P<0.05)。电镜下模型家兔见不同程度线粒体肿胀、肌质网扩张。结论酸和胆汁混合灌注可以成功制备家兔混合反流性食管炎动物模型。
Objective To prepare animal model of mixed reflux esophagitis by mixed infusion of acid and bile. Methods Rabbit mixed reflux esophagitis model was established by mixed infusion of hydrochloric acid and porcine bile. The model group was divided into 4 d group, 7 d group and 10 d group at different times of perfusion. Normal group was infused with normal saline. The changes of esophagus in each group were observed under light microscope and electron microscope, and the grading was compared. Results After mixed infusion pathological changes of the lower esophagus obvious. There were varying degrees of mucosal roughness, hyperemia and submucosal hyperplasia in the lower esophagus of the three groups of rabbits in the model group, scattered or extensive erosion, and ulcers with different degrees were observed. There were significant differences in the gross esophagogastric scores between the three groups (P <0.01). Model rabbit showed part of squamous epithelium shrinkage, edema eosinophils, neutrophil infiltration, submucosa vasodilation, congestion, see varying degrees of ulcers, tissue necrosis, esophageal muscle layer also see varying degrees of inflammation Cell infiltration. Three groups of rabbits esophageal pathological differences were statistically significant (P <0.05). Under electron microscope, the rabbits showed mitochondria swelling and sarcoplasmic reticulum swelling. Conclusion The mixed bile acid and bile can be successfully prepared in rabbits mixed reflux esophagitis animal model.
