目的 了解血管内皮生长因子(VEGF)在新生鼠缺氧缺血性脑损伤后表达的动态变化。方法 采用Rice法制备缺氧缺血性脑病模型,将80只7日龄SD大鼠随机分为:正常对照组、模型后即刻组、1h组、3h组、6h组、12h组、24h组、48h组、72h组、7d组。用免疫组织化学方法观察VEGF表达的动态变化。结果 正常对照组中大鼠脑内几乎无VEGF表达;模型组结扎侧大脑皮质,纹状体,海马在模型后即刻即有VEGF表达,3~12h达高峰,随后开始下降,7d后呈低水平稳定表达。结论缺氧缺血作为一种信号刺激VEGF在神经细胞内表达增加;VEGF的表达增加具有时间依赖性;VEGF对缺氧缺血性神经元损伤可能具有一定的保护作用。
Objective To investigate the dynamic changes of vascular endothelial growth factor (VEGF) expression in neonatal rats after hypoxic-ischemic brain damage. Methods The model of hypoxic-ischemic encephalopathy was prepared by the method of Rice. Eighty SD rats of 7 days old were randomly divided into normal control group, immediate after model group, 1h group, 3h group, 6h group, 12h group, 24h group, 48h group, 72h group, 7d group. The dynamic changes of VEGF expression were observed by immunohistochemistry. Results There was almost no VEGF expression in the normal control group. The expression of VEGF in the cortex, striatum and hippocampus of the model group immediately after the model reached the peak at 3 ~ 12h, then began to decline, and then decreased at 7d Stable expression. Conclusion Hypoxia and ischemia as a signal stimulates the expression of VEGF in neurons increased; VEGF expression increased in a time-dependent manner; VEGF may have a protective effect on hypoxic-ischemic neuronal damage.
