目的:探讨Xk1p2靶蛋白(targeting protein for Xk1p2,TPX2)和胸苷磷酸化酶(thymidine phosphoryl-ase,T P)在胃腺癌中的表达及其与临床病理因素的关系。方法:应用免疫组织化学SP法,检测75例胃腺癌(胃腺癌组)、33例异型增生(癌旁异型增生组)和75例正常胃黏膜组织(正常胃黏膜组)中TPX2和TP蛋白的表达。结果:胃腺癌组TPX2和TP的阳性表达率分别为70.7%和62.7%;癌旁异型增生组为33.3%和39.4%;正常胃黏膜组为2.7%和5.3%。TPX2和TP在正常胃黏膜组、癌旁异型增生组及胃腺癌组中的表达率依次增高,经多个样本率的比较,3组间均有统计学意义(P<0.01)。TPX2蛋白的表达与肿瘤的分级、分期、浸润深度及淋巴结转移有关(P<0.05),与年龄、肿瘤大小无关(P>0.05)。TP的阳性表达率与肿瘤的分期、浸润深度、淋巴结转移及肿瘤大小均有统计学意义(P<0.05),与年龄、分化程度无关(P>0.05)。TPX2与TP在胃腺癌中的表达呈正相关关系(r=0.4697,P<0.01)。结论:TPX2和TP可能共同参与了胃腺癌的发生、发展及转移。联合检测TPX2和T P有望成为胃腺癌的早期诊断及预后判断的候选标志物。
Objective: To investigate the expression of Xk1p2 target protein (TPX2) and thymidine phosphoryl-ase (T P) in gastric adenocarcinoma and its relationship with clinicopathological factors. Methods: The expressions of TPX2 and TP in 75 cases of gastric adenocarcinoma (gastric adenocarcinoma), 33 cases of dysplasia (paracancerous dysplasia) and 75 cases of normal gastric mucosa (normal gastric mucosa) were detected by immunohistochemical SP method expression. Results: The positive rates of TPX2 and TP in gastric adenocarcinoma were 70.7% and 62.7% respectively, while those in paracarcinomatous dysplasia were 33.3% and 39.4%. The normal gastric mucosa were 2.7% and 5.3% respectively. The expression rates of TPX2 and TP in the normal gastric mucosa group, paracancerous dysplasia group and gastric adenocarcinoma group were successively higher than those in the normal gastric mucosa group and the gastric adenocarcinoma group. There were significant differences among the three groups (P <0.01). The expression of TPX2 protein was correlated with tumor grade, stage, depth of invasion and lymph node metastasis (P <0.05), but not with age and tumor size (P> 0.05). The positive rate of TP was correlated with tumor stage, depth of invasion, lymph node metastasis and tumor size (P <0.05), but not with age and differentiation (P> 0.05). The expression of TPX2 and TP in gastric adenocarcinoma was positively correlated (r = 0.4697, P <0.01). Conclusion: TPX2 and TP may participate in the occurrence, development and metastasis of gastric adenocarcinoma. Combined detection of TPX2 and Tp is expected to be a candidate marker for early diagnosis and prognosis of gastric adenocarcinoma.
