观察了低剂量辐射对人脐血 T 淋巴细胞膜分子表达的影响。应用 62 mGy γ射线照射新鲜分离的脐血淋巴细胞,照前及照后 4 h,12 h,24 h 分别采用单克隆抗体直接免疫荧光标记流式细胞术检测TCR,CD3,CD4,CD8 分子的表达。实验结果表明低剂量辐射后,脐血淋巴细胞 CD3,TCR/CD3 复合物,CD4,CD8 分子表达显著上调,且均在 4~24 h 内随时间推移而逐步增幅;CD4/CD8 比值无显著性变化。以上结果提示:低剂量辐射可促进脐血 T 淋巴细胞的成熟、活化和信号的转导,从而可能在脐血移植中加速免疫功能重建,增强 GVL 效应,降低肿瘤的复发率。
The effect of low dose radiation on the molecular expression of T lymphocyte in human umbilical cord blood was observed. Freshly isolated umbilical cord blood lymphocytes were irradiated with 62 mGy γ-rays and the expression of TCR, CD3, CD4 and CD8 were detected by direct immunofluorescence labeling with monoclonal antibody at 4 h, 12 h and 24 h after irradiation expression. The results showed that the expressions of CD3, TCR / CD3 complex, CD4 and CD8 in cord blood lymphocytes were significantly increased after low dose radiation, and all of them were increased gradually with the passage of time within 4 ~ 24 h; the ratio of CD4 / CD8 was not significant Variety. The above results suggest that low dose radiation can promote the maturation, activation and signal transduction of T lymphocytes in cord blood, which may accelerate the immune function reconstruction in cord blood transplantation, enhance GVL effect and reduce the recurrence rate of tumor.
