目的观察卵清蛋白(OVA)诱导建立的急性支气管哮喘大鼠模型,采用外源性硫氢化钠(NaHS)处理后Th1/Th2细胞因子及肺部黏蛋白基因Muc5ac的表达情况,探讨气体信号分子硫化氢(H2S)在哮喘发病中的作用。方法26只健康SD大鼠按照随机数字表法分为正常对照组(A组)、哮喘组(B组)和NaHS干预组(C组),每组分别为8只、9只、9只。激发24h后解剖大鼠,取腹腔静脉血检测其IL-4及IFN水平。取肺组织观察大鼠支气管周围的炎性细胞浸润程度并进行评分;采用免疫组织化学染色法测定大鼠肺中Muc5ac的表达情况。结果光镜下哮喘大鼠支气管周围炎性细胞浸润程度评分(采用中位数表示)A组1分,B组4分,C组2分,3组间比较差异有统计学意义(H=13.75P<0.05)。A、B、C组血浆IL-4水平分别为(99.25±31.60)、(185.40±139.30)和(262.00±27.10)pg/L,3组间比较差异有统计学意义(F=7.706P<0.05)。A、B、C组IFN-γ水平分别为(177.2±73.0)、(91.8±32.3)和(120.7±31.5)pg/L,3组间比较差异有统计学意义(F=6.731P<0.05)。A、B、C组肺Muc5ac蛋白水平(用阳性面积表示)分别为(501.75±270.58)、(1330.22±662.87)和(715.88±282.49)μm2,3组间比较差异有统计学意义(F=11.82P<0.01)。结论哮喘大鼠黏蛋白基因Muc5ac表达增加,外源性H2S可减轻哮喘呼吸道炎性反应,对哮喘急性发作起保护作用。
Objective To observe the expression of Th1 / Th2 cytokines and mucin gene Muc5ac in acute asthmatic rat induced by ovalbumin (OVA) and the treatment of exogenous sodium hydrosulfide (NaHS) Hydrogen sulfide (H2S) in the pathogenesis of asthma. Methods Twenty-six healthy SD rats were divided into normal control group (group A), asthma group (group B) and NaHS intervention group (group C) according to the random number table. Each group was 8, 9 and 9 respectively. Rats were dissected 24h after anoxia, and the levels of IL-4 and IFN were measured by abdominal venous blood. The degree of infiltration of inflammatory cells around the bronchus in the lungs was observed and scored. The expression of Muc5ac in rat lungs was detected by immunohistochemical staining. Results The score of bronchial inflammatory cell infiltration in asthmatic rats under light microscope (median) was 1 in group A, 4 in group B, and 2 in group C, with significant difference between the three groups (H = 13.75 P <0.05). The levels of plasma IL-4 in groups A, B and C were (99.25 ± 31.60), (185.40 ± 139.30) and (262.00 ± 27.10) pg / L, respectively. There were significant differences among the three groups (F = 7.706P <0.05 ). The levels of IFN-γ in group A, B and C were (177.2 ± 73.0), (91.8 ± 32.3) and (120.7 ± 31.5) pg / L, respectively. There were significant differences among the three groups (F = 6.731P <0.05) . The Muc5ac protein levels of lung in group A, B and C were (501.75 ± 270.58), (1330.22 ± 662.87) and (715.88 ± 282.49) μm2, respectively, with significant difference between the three groups (F = 11.82 P <0.01). Conclusions Muc5ac expression is increased in asthmatic rats. Exogenous H2S can reduce the inflammatory reaction of asthmatic airways and protect the acute asthma attack.
