目的研究路边青与穿心莲配伍使用对脱水穿心莲内酯(DAP)的小鼠血液药动学性质的影响。方法小鼠灌胃穿心莲总内酯干浸膏和复方穿心莲干浸膏药液后于不同时间点取血,血样经前处理后用反相高效液相色谱分离、测定脱水穿心莲内酯含量,采用3P97药动学软件对数据进行处理。结果小鼠灌胃穿心莲总内酯干浸膏和复方穿心莲片干浸膏后,脱水穿心莲内酯的血药浓度动态变化规律均符合一室开放模型。路边青与穿心莲配伍使用可导致脱水穿心莲内酯的吸收慢、消除快、生物利用度低。结论穿心莲与路边青配伍使用是否科学,值得深入研究。
Objective To study the effect of compatibility of Rupinqing and Andrographis on blood pharmacokinetics of dehydroandrographolide (DAP) in mice. Methods The mice were orally administered with Andrographis paniculata extract and Compound Andrographis paniculata extract liquid. The blood samples were taken at different time points. The blood samples were pre-treated and separated by reversed-phase high-performance liquid chromatography to determine the content of dehydroandrographolide, using 3P97. Pharmacokinetic software processes the data. RESULTS: After mice were intragastrically administered with total extract of Andrographis paniculata and compound dry heart extract, the dynamic changes of blood concentration of dehydroandrographolide were consistent with one-compartment open model. The use of Roadside Green in combination with Andrographis may result in slow absorption, rapid elimination, and low bioavailability of dehydroandrographolide. Conclusion The compatibility of Andrographis paniculata with roadside green is scientific and worthy of further study.
