目的了解辽宁省丹东市鸭绿江沿岸虫媒病毒的种类和分布情况。方法使用诱蚊灯捕蚊,通过组织培养法分离病毒,并对病毒分离物进行血清学和分子生物学鉴定。结果 2007年7月在辽宁省丹东市鸭绿江沿岸6个采集点采集到3属3种共3359只蚊虫标本,从中分离到11株病毒分离物,鉴定结果显示有5株为版纳病毒,3株为环状病毒样病毒,其余3株分离物有待进一步鉴定;版纳病毒新分离株序列分析结果显示,5株版纳病毒之间第12片段序列高度同源,核苷酸同源性为92.1%~99.8%;氨基酸同源性为92.8%~99.5%;并且与北京分离株和云南分离株同源性较高,核苷酸同源性为91.3%~98.7%;氨基酸同源性为90.8%~100%;系统进化分析提示,版纳病毒新分离株与北京、云南分离株位于同一个进化分支中。结论在辽宁省丹东市鸭绿江沿岸首次分离到版纳病毒和环状病毒样病毒,新分离版纳病毒与北京、云南分离株进化关系较近。
Objective To understand the species and distribution of arboviruses along the Yalu River in Dandong City, Liaoning Province. Methods The mosquito lamp was used to catch mosquitoes and the virus was isolated by tissue culture method. The virus isolates were identified by serology and molecular biology. Results A total of 3359 mosquitoes from 3 genera and 3 genuses were collected from 6 collection sites along the Yalu River in Dandong City, Liaoning Province in July 2007, and 11 virus isolates were isolated from them. The identification results showed that 5 were of the genus Banna virus and 3 were The other three isolates had to be further identified. Sequence analysis of the new version of the BAVV showed that the sequence of the 12th fragment of the five BAVs was highly homologous with nucleotide homology of 92.1% ~ 99.8 %; The amino acid homology was 92.8% -99.5%; And the nucleotide homology was 91.3% -98.7% with Beijing and Yunnan isolates, the amino acid homology was 90.8% -100 %. Phylogenetic analysis suggested that the new isolates of BAVs were located in the same evolutionary branch with Beijing and Yunnan isolates. Conclusion For the first time, the isolates of Banna virus and Rattus norvegicus were isolated along the Yalu River in Dandong City, Liaoning Province. The new isolated Banna virus was closely related to the isolates of Beijing and Yunnan.
